ABSTRACT

Exosomes play important roles in cell communication and information transmission.miRNA-126 carried by mesenchymal stem cells exosomes can reduce inflammatory response and protect vascular endothelium by regulating protein expression, thus blocking or alleviating cardiac dysfunction during septic shock, which is expected to become a new way to treat septic shock cardiac dysfunction.

ABSTRACT

Objective:To investigate the effect of miRNA-126 on proliferation, apoptosis and migration of thyroid cancer SW579 cells, and further to explore its mechanism.Methods:Thyroid cancer SW579 cells were cultured in vitro, and the expression of miRNA-126 in normal thyroid Nthy-ori3-1 cells and SW579 cells was detected by qPCR. The cells were divided into blank control group (without any treatment) , experimental group (transfected with miRNA-126 expression plasmid) and negative control group (transfected with empty plasmid vector) . SW579 cells with overexpression of miRNA-126 were constructed by transfection of plasmids. CCK-8 test was used to detect cell proliferation; Transwell cell test was used to detect cell migration and invasion; Flow cytometry was employed to detect apoptosis and changes in reactive oxygen levels; Western blot was used to detect Notch-1/Akt pathway related protein expression.Results:The expression level of miRNA-126 in SW579 cells was 0.25±0.07, and the difference was statistically significant compared with that in Nthy-ori3-1 cells (P<0.001) . The cell survival rates of blank control group, experimental group and negative control group were (105.70 ± 7.61) , (98.60 ± 5.42) and (62.70 ± 3.82) ; The apoptosis rates were (9.14 ± 0.83) , (12.28 ± 1.34) and (36.39 ± 3.21) (all P < 0.05) ; The cell migration rates were (34.51 ± 2.45) , (33.29 ± 3.17) and (11.22 ± 1.23) (all P < 0.05) ; The levels of ROS were (1.02 ± 0.07) , (1.08 ± 0.11) and (6.54 ± 0.74) (all P < 0.05) .Conclusion:Overexpression of miR-NA-126 can inhibit Notch-1/Akt pathway by up-regulating intracellular reactive oxygen species, inhibit the proliferation and migration of SW579 cells, and induce apoptosis in SW579 cells, which provides a certain basis for the study of miRNA-126 in thyroid cancer.

ABSTRACT

ObjectiveThe mechanism of cerebral ischemia-reperfusion injury is highly associated with the inflammatory response. MiRNA-126 plays a key role in vascular inflammation. This study aims to investigate the effect of miRNA-126 on the inflammatory response in mice accompanying cerebral ischemia-reperfusion injury through the NLRP3/NF-κB signal axis, and to explore the mechanisms involved.Methods A total of forty-eight male mice were randomly divided into four groups: the sham-operated group, cerebral ischemia-reperfusion model group, miRNA negative control group (miRNA-126 agomir NC group) and miRNA-126 overexpression group (miRNA-126 agomir group), and each group included twelve mice. The neurobehavioral score was recorded. The left-brain of the mice was sacrificed after anesthesia, and the water content of the brain tissue was measured. HE staining and light microscopy were used to identify the histopathological changes of the cerebral of the mice. The expression levels of inflammatory cytokines IL-1β and IL-6 in brain tissue and serum of mice were detected by ELISA. Western Blot method was used to determine the protein expression levels of NLRP3, ASC, caspase-1, NF-κB p65, NF-κB P50, p-NF-κB 65 and p-NF-κB 50 in brain tissues of mice in each group. RT-PCR was used to test the expression levels of miRNA126, NLRP3, ASC, caspase-1, NF-κB p65 and NF-κB P50 in brain tissue and serum of mice.ResultsIn the sham-operated group, the morphology, and structure of cerebral cortex were normal as healthy mice, being with the dense and orderly arrangement of nerve fibers, with no occurrence of impaired nerve function, and the neurobehavioral score was zero. In both of model group and the miRNA-126 agomir NC group, the ruptured cerebral cortex could be observed visually being with necrotic and disordered cells. The blurred pyknosis and interstitial edema occurred with increased water content of brain tissue. The nerve damage was observed with a significantly increased neurobehavioral score (P<0.05). Compared to the model group, the pathological morphology of the cerebral cortex in the miRNA-126 agomir group was significantly improved, and the number of necrotic cells was decreased, the arrangement of which was denser and more orderly. Reduced interstitial edema and the neurobehavioral score were identified. The significantly improved nerve injury and the decreased water content of brain tissue were observed as well (P<0.05). Compared to the sham-operated group A, the expression level of miRNA-126 mRNA in the model group and the miRNA-126 agomir NC group decreased significantly. The expression level of IL-1β and IL-6 increased, while the expression levels of NLRP3, ASC, caspase-1, NF-κB p65, NF-κB p50, p-NF-κB p65, p-NF-κB p50, and NLRP3, ASC, caspase-1, NF-κB p65, NF-κB p50 mRNA increased generally (P<0.05). Compared to the model group and the miRNA-126 agomir NC group, the expression level of miRNA-126 mRNA in the miRNA-126 agomir group increased. However, the expression level of IL-1β and IL-6 decreased, and the expression level of NLRP3/NF-κB signal axis related gene protein and mRNA decreased (P<0.05).ConclusionOverexpression of miRNA-126 can inhibit the expression of NLRP3/NF-κB signal axis related genes and the level of inflammation in brain tissue, and improve the neurological injury of cerebral ischemia-reperfusion in mice.

ABSTRACT

Objective@#To investigate the expressions of serum miRNA-126 (miR-126) and miRNA-30c (miR-30c) in patients with pancreatic cancer, and to analyze the relationship with the occurrence of pancreatic cancer as well as the diagnostic value.@*Methods@#A total of 110 patients with pancreatic cancer diagnosed at the 928th Hospital of the Joint Service Support Force of PLA from January 2014 to December 2018 were selected, and 110 healthy people were also selected as the control group. The expression levels of serum miR-126 and miR-30c of 110 patients and the healthy controls were detected by using real-time quantitative polymerase chain reaction (qRT-PCR), and their relationship with clinicopathological features of pancreatic cancer was analyzed.@*Results@#The levels of serum miR-126 and miR-30c in pancreatic cancer group were lower than those in the healthy control group (0.43±0.12 vs. 1.02±0.27, t = 19.394, P < 0.01; 0.52±0.17 vs. 1.04±0.31, t = 15.426, P < 0.01). There was a positive correlation between levels of serum miR-126 and miR-30c in patients with pancreatic cancer (r = 0.399, P < 0.01). Expression levels of serum miR-126 and miR-30c were not correlated with age, gender and body mass index (BMI) of pancreatic cancer patients (all P > 0.05), but related with tumor size, differentiation degree, TNM stage and lymph node metastasis (all P < 0.05). High expression levels of serum miR-126 and miR-30c were protective factors for pancreatic cancer (all P < 0.05), while smoking, drinking and gallstones were risk factors for pancreatic cancer (all P < 0.05). The area under the curve of combined detection of serum miR-126 and miR-30c for diagnosis of pancreatic cancer was 0.906, the sensitivity was 90.80%, and the specificity was 82.20%.@*Conclusions@#The expression levels of serum miR-126 and miR-30c in patients with pancreatic cancer are low. Both of them may be involved in the occurrence and development of pancreatic cancer, and may be used as early diagnostic markers of pancreatic cancer.

ABSTRACT

Objective To study the clinical application of serum miRNA 21 and miRNA-126 in diagnosis of the coronary artery restenosis after percutaneous coronary intervention (PCI).Methods The serum and clinical data of 82 cases of acute coronary syndrome (ACS) patients treated with PCI from the Hospital of Jingyang Country and the Affiliated Hospital of Shaanxi University of Chinese Traditional Medicine (stenosis group consisted of 30 cases,non-stenosis group consisted of 52 cases),and healthy persons (control group consisted of 33 cases) were collected.The relative expression levels of serum miRNA-21 and miRNA 126 were detected using real-time qPCR technology in the comparative analysis for their changes in the groups.Intravascular ultrasound (IVUS) was used to evaluate the occurrence of restenosis after PCI.Results Serum miRNA-21 and miRNA 126 levels in the control group,the non-stenosis group and the stenosis group were,,(0.22 ± 0.03,0.43±0.08,0.92±0.17) and (0.87±0.15,0.49±0.10,0.23±0.04),respectively.Compared with the control group,the expression level of serum miRNA-21 in the stenosis group and the non stenosis group were significantly increased,while the miRNA-126 levels was significantly decreased (P=0.000),the differences were statistically significant.Compared with the non stenosis group,the level of miRNA-21 in the stenosis group was significantly increased,while the level of miRNA-126 was significantly decreased (P=0.000),the differences were statistically significant.The results of IVUS showed that the plaque area (PLA) of the patients in the stenosis group was significantly higher than that in the non stenosis group,while the minimal luman area (MLA) was significantly decreased,and the differences were statistically significant (P=0.000).There was a negative correlation between the expression of two markers in the stenosis group (r=-0.919,P<0.01).The levels of miRNA-126 and miRNA 21 in the stenosis group were correlated with PLA and MLA (r=-0.945,0.926,P< 0.01) and (r=0.933,-0.917,P<0.01).Conclusion The detection of serum miRNA-21 and miRNA-126 levels can be used in diagnosis and prediction for the coronary artary restenosis of ACS after PCI,as the gene detection index of the disease.

ABSTRACT

Objective To assess the influence of miRNA (miR)-124 ,miR-26A and miR-126 single nucleotide polymorphism (SNPs) on the prognosis in patients with colorectal cancer (CRC) .Methods A total of 724 cases of patients with CRC treated in this hospital from July 2011 to December 2013 served as the research subjects .The peripheral blood samples were collected ,and genotype analysis of miR-124 rs531564 ,miR-26A rs7372209 and miR-126 rs4636297 SNPs locus were performed by using the PCR-LDR .The patient′s overall survival (OS) and progression-free survival (PFS) were calculated by using Kaplan-Meier method ,and the univariate log-rank method was used to analyse the influence of different genotypes and haplotypes of SNPs on OS and PFS .Re-sults The OS after operation in CRC patient′s carrying miR-124 rs531564 CG genotype was decreased compared with that in pa-tients carrying CC genotype ,and the patient′s death risk was increased[HR2 =1 .450 ,95% CI(1 .043-2 .017) ,P=0 .026];the pa-tient′s death risk in patients carrying GG genotype was also increased compared with that in patients carrying CC genotype [HR2 =2 .339 ,95% CI(1 .171-4 .674)];in the dominant gene model ,the OS in patients carrying CG +GG genotype was significantly de-creased compared with patients carrying CC genotype [HR2 = 1 .532 ,95% CI(1 .119 -2 .096) ,P= 0 .008];in the recessive gene model ,no statistically significant difference was found in OS between patients carrying GG genotype and those carrying CC + CG genotype[HR2 =1 .975 ,95% CI(1 .000-3 .900) ,P=0 .0052];in the co-dominant model ,compared with patients carrying CC+CG genotype ,the OS in patients carrying CG genotype was decreased and their death risk was increased [HR2 =1 .395 ,95% CI(1 .007-1 .931)] .There was no correlation of miR-26A rs7372209 locus and miR-126 rs4636297 locus gene SNPS to OS and PFS in CRC patients (P>0 .05) .Conclusion miR-124 rs531564 gene polymorphism might be an independent influence factor of the prognosis in CRC patients .

ABSTRACT

Objective To investigate the clinical application of serum miRNA-126,miRNA-155 detection in evaluation of plaque property in the carotid atherosclerotic(CAS)desease.Methods A total of 75 patients with the CAS from May 2015 to May 2017 in the Xianyang Central Hospital and Shiquan Country Chinese Traditional Medicine was chosen,consisted of 35 cases of vulvernable plaque group and 40 cases of stable plaque group.Meanwhile,39 cases of healthy physical examines at the same time were regarded as the control group.The expression levels of serum miRNA-126,miRNA-155 in the groups were detected using the real-time reverse transcription-polymerase chain reaction technique.The largest carotid artery plaque thickness(MAPT)and intima-media thickness(IMT)in the groups were measured using the cervical enhancement CT.Re-sults The results of MAPT and IMT were(3.27±1.01 mm,1.93±0.51 mm)in the vulvernable plaque group and(2.50 ±0.79 mm,1.60±0.26 mm)in the stable plaque group.The carotid artery largest plaque thickness and intima-media thick-ness was higher in the vulvernable plaque group than in the stable plaque group(t=9.76,7.86,P<0.01),and there were significant differenes between the two groups.The expression levels of serum miRNA-126and miRNA-155 were(0.22 ± 0.06,0.87±0.18)in the vulvernable plaque group,(0.50±0.12,0.47±0.10)in the stable plaque group and(0.90±0.19, 0.19±0.05)in the control group.MiRNA-155 expression levels significantly increased in stable plaque group and vulvern-able plaque group compared with in the control group,which increased in the vulvernable plaque group compared with in the stable plaque group,and miRNA-126 expression levels markedly decreased,the differences were statistically significant(F=119.3,102.9,P<0.01).In the vulvernable plaque group,miRNA-126 expression negatively correlated with miRNA-155(r=0.912,P<0.01).miRNA-126 expression levels were inversely associated with the carotid artery largest plaque thickness and the intima-media thickness(r=-0.913,-0.893,P<0.01).While miRNA-155 expression levels were positively corre-lated with them(r=0.899,0.907,P<0.01).Conclusion Serum miRNA-155,miRNA-126 detection can be applied to pre-diction of CAS plaques rupture,and may become a useful warning marker of ischemic stroke events.

ABSTRACT

Objective To explore the mechanism of plasma circulating miRNA-126 and miRNA-28-3p in diabetes mellitus (DM) patients,and to identify the related bioinformatics analysis.Methods Randomly selected 80 DM patients as the observation group and 80 non-DM patients as the control group.The plasma circulating miRNA-126 and miRNA-28-3p were analyzed by qRT-PCR,and its target genes,biological information,related lncRNA and circRNA were predicted.Results The circulating miRNA-126(0.115 0±0.014 4 vs.0.0019±0.000 6) and miRNA-28-3p(0.1386±0.01724 vs.0.000 6±0.000 05) levels in the observation group were significantly higher than those in the control group,and the differences were statistically significant (P<0.01).Pearson correlation coefficient of miRNA-126 and miRNA-28-3p was 0.433 5(P<0.01).ROC curve analysis of miRNA-126 and miRNA-28-3p showed that the differences of the area under curve were statistically significant between the two groups (P<0.01).Bioinformatics prediction showed that miRNA-126 and miRNA-28-3p may be involved in regulation of the insulin signaling pathway,insulin receptor signaling pathway,insulin/insulin growth factor signaling pathway,mitogen-activated protein kinase (MAPK) signaling pathway and angiogenesis.And it may be associated with a variety of lncRNA and circRNA.Conclusion Circulating miRNA-126 and miRNA-28-3p can be a novel biomarker of DM as it may participate in the mechanism of DM by regulating insulin and insulin growth factor related signaling pathways and be associated with some related lncRNA and circRNA.

miRNA is a class of small non-coding RNAS miRNA-126 is abnormally expressed in many tumors,especially in non-small cell lung cancer,by acting on different target genes and signaling pathways,which can influence the behaviors of tumor cells, such as growth,proliferation,invasion and metastasis. Therefore,study of miRNA-126 might provide a novel direction for clarifying the pathogenesis,diagnosis and treatment of lung cancer.

ABSTRACT

Objective:To observe expression level change of miRNA?126 in the myocardial tissue of rats with myocardial infarction and to observe the influence of Shuxuetong injection. Methods:Myocardial infarction model was established in 80 SD rats, 60 surviv?al rats were randomly divided into Shuxuetong group and model group, the other 30 rats were randomly selected as control group. After the success of the modeling, Shuxuetong group received Shuxuetong injection treatment, model group and control group received same amount of normal saline. Real?time quantitative PCR was used to detect miRNA?126 expression levels in rat myocardial tissue at dife?rent time after operation. Results: After 7 days and 14 days, miRNA?126 expression levels in rats myocardial tissue in Shuxuetong group and model group were significantly lower than that in the control group ( P<0?05) , but there was no difference between the two groups. After 14 days, miRNA?126 expression levels in the Shuxuetong group were higher than that after 7 days ( P<0?05) . Conclu?sions:miRNA?126 in 7 days after myocardial infarction significantly reduced. With the passing of time, miRNA?126 expression levels increased gradually and Shuxuetong can increase the expression levels of miRNA?126 .

ABSTRACT

Objective To investigate the corelation between miRNA-126-3p and pathogenesis of pulmonary hypertension . Methods Totally 25 patients with congenital heart disease ,including 11 cases with pulmonary hypertension and 14 control cases , were recruited .The expression of miRNA-126-3p was detected by qRT-PCR ,and we used starBase to predict the biological informa-tion miRNA-126-3p ,and then verified it by the level of mRNA and protein .Results There was no statistical difference in age ,gen-der ,and biochemical examination(P> 0 .05) ;the expression of miRNA-126-3p in the pulmonary hypertension cases was significantly higher(P< 0 .01) ;starBase found that miRNA-126-3p may participate in binding protein ,signal transduction ,cell differentiation and regulation of cell morphology ,the regulation of MAPK and insulin receptor signaling pathways ,etc .Its target genes mainly contain VEGFA ,SPRED1 ,PIK3R2 ,etc ;the mRNA and protein level of VEGFA in PH group were statistically significantly than control group(P< 0 .01) ;the miRNA-126-3p and VEGFA showed positive correlation(P< 0 .01) .Conclusion miRNA-126-3p may partici-pate in the occurrence of pulmonary hypertension by regulating VEGFA .
